The present invention provides a method for determining the presence of Mycobacterium avium subspecies paratuberculosis (MAP) in a sample. The method involves using a pair of oligonucleotide probes and detecting the presence or absence of fluorescence resonance energy transfer (FRET) between the donor fluorescent moiety of the first probe and the corresponding acceptor fluorescent moiety of the second probe. The present invention also provides a method for isolating and/or extracting DNA from a microorganism.

L'invention concerne un procédé destiné à déterminer la présence de la sous-espèce paratuberculosis du Mycobacterium avium (MAP) dans un échantillon. Le procédé consiste à utiliser une paire de sondes oligonucléotides et à détecter la présence ou l'absence de transfert d'énergie par résonance de fluorescence (FRET) entre la fraction fluorescente donneuse de la première sonde et dans la fraction fluorescente acceptrice correspondante de la seconde sonde. Cette invention concerne également un procédé d'isolation et/ou d'extraction d'ADN à partir d'un micro-organisme.
