An improved kinetic assay is provided for spectrophotometrically determining dissolved carbon dioxide (CO2) in body fluid wherein at pH 8.0 the CO2 is present substantially as bicarbonate ion (HCO3). The sample is subjected to a reaction mixture containing phosphoenolpyruvate (PEP) and phosphoenolpyruvate carboxylase (PEPC) which enzymatically catalyzes conversion of HCO3 to oxaloacetate (OA). The OA produced is subject to a second reaction mixture containing NADH and malate dehydrogenase (MDH) which reduces the OA to malate and oxidizes the NADH to NAD +. The CO2 level in the sample may be indirectly determined by measuring the change in NADH concentration. An inhibitor is added to the reaction mixture which renders the reaction kinetics of the system essentially first order over the entire bicarbonate concentration range of interest. Best results have been obtained through use of an inhibitor which provides an anion selected from the group consisting of C10 3, C10 4, SCN, or I.

Un essai cinétique amélioré est permis par dosage spectrophotométrique du dioxyde de carbone dissous (CO2) dans les fluides corporels à pH 8,0, pH auquel le CO2 est substantiellement présent sous forme d'ion bicarbonate (HCO3-). L'échantillon est soumis à un mélange réactionnel contenant du phosphoénolpyruvate (PEP) et de la phosphoénolpyruvate-carboxylase (PEPC) qui, par catalyse enzymatique, transforme le HCO3- en oxaloacétate (OA). L'OA produit est soumis à un second mélange réactionnel contenant de la NADH et de la malate-désyhdrogénase (MDH) qui réduit l'OA en malate et oxyde la NADH en NAD+. La teneur en CO2 de l'échantillon peut être déterminée indirectement par mesure du changement de la teneur en NADH. Un inhibiteur est ajouté au mélange réactionnel qui rend la cinétique de réaction du système essentiellement d'ordre 1 sur toute la plage d'intérêt de teneur en bicarbonate. Les meilleurs résultats ont été obtenus par utilisation d'un inhibiteur qui fournit un anion choisi parmi les suivants : ClO3, ClO4, SCN ou I.
