A method for purifying thrombin substrates and/or inhibitors characterized by involving the step of effecting affinity chromatography with the use of an affinity adsorbent having thrombin or its analogue from which the activity (nucleophilicity) has been taken off without inducing any steric hindrance in binding, as a ligand, to the substrates and/or inhibitors. This method aims at efficiently, safely and stably purifying inexpensive and highly pure thrombin substrates and/or inhibitors by using a novel and inexpensive separation/purification means with the use of a convenient separation/purification procedure and a small number of purification/separation steps. By using the above method, blood coagulation factors VIII and XIII, protein C, etc. can be purified at a high purity in a high yield as compared with the conventional purification methods.
L'invention concerne un procédé de purification de substrats de thrombine et/ou d'inhibiteurs de thrombine, caractérisé par l'étape consistant à effectuer une chromatographie d'affinité au moyen d'un adsorbant d'affinité contenant une thrombine ou un analogue de celle-ci dont on a supprimé l'activité (nucléophile) sans provoquer d'encombrement stérique dans la capacité de liaison, comme ligand, à des substrats et/ou des inhibiteurs. Le procédé vise à mettre en oeuvre une purification efficace, sûre et stable de substrats de thrombine et/ou d'inhibiteurs de thrombine bon marché et de grande pureté, par un nouveau moyen de séparation/purification utilisant une procédure adéquate de séparation/purification et comportant un petit nombre d'étapes de purification/séparation. L'utilisation du procédé mentionné permet d'obtenir des facteurs de coagulation VIII et XIII, de protéine C etc. de grande pureté et d'assurer des rendements élevés par comparaison avec des procédés de purification classiques.
