Molecular cloning and expression of a human gene encoding a polypeptide activator of proteasomes is disclosed. The expressed activator has an Mr of about 29,000 and is functional in activating proteasomes in vitro. In vivo this activator polypeptide is inducible with .gamma.-interferon in HeLa cells and occurs with a non-.gamma.-interferon-inducible polypeptide with an Mr of about 31,000 in a hexameric activator complex. The activator protein contains a lysine and glutamate rich region termed a KEKE motif. The KEKE motif appears to promote association between proteins and selection of peptides for presentation on MHC Class I receptors. A method for enhancing cell-mediated immunity against or tolerance to a selected immunogenic peptide is described comprising expressing activator and the selected peptide, wherein the selected peptide is adjacent to a KEKE motif, in an appropriate cell.
On décrit le clonage moléculaire et l'expression d'un gène humain codant pour un activateur polypeptidique des protéasomes. L'activateur exprimé a une valeur de M¿r? d'environ 29000 et il agit pour activer les protéasomes in vitro. in vivo, cet activateur polypeptidique est inductible par l'interféron gamma dans les cellules HeLa et il est accompagné d'un polypeptide non inductible par l'interféron gamma, avec une valeur de M¿r? de 31000 environ, sous la forme d'un complexe activateur hexamère. L'activateur protéinique contient une région riche en lysine et en glutamate appelée molit KEKE. Le molit KEKE semble favoriser l'association entre les protéines et la sélection de peptides pour présentation sur les récepteurs de classe I du complexe majeur d'histocompatibilité. On décrit une méthode pour améliorer l'immunité provoquée par des cellules contre un peptide immunogène sélectionné ou la tolérance vis-à-vis de celui-ci. Cette méthode consiste à exprimer dans une cellule appropriée l'activateur et le peptide sélectionné, ce dernier jouxtant le molit KEKE.
