The present invention relates to a method for screening compounds to identify compounds that bind to a specific target, comprising the steps of: (a) providing a target immobilized to a solid support; (b) generating a first NMR spectrum of the said compounds to be screened in the presence of the solid support without the target molecule immobilized thereto; (c) generating a second NMR spectrum of the said compounds to be screened in the presence of the solid support with the target molecule immobilized thereto; and (d) comparing said first and second NMR spectrum to determine differences between said first and second NMR spectrum. Further the invention relates to an NMR apparatus having arranged therein a particular NMR probe.
La présente invention concerne un procédé de recherche systématique de composés permettant d'identifier des composés se liant à une cible spécifique. A cet effet, on commence par (a) se procurer une cible immobilisée sur un support solide, puis (b) on génère un premier spectre RMN des composés à soumettre à recherche systématique en présence du support solide sans que la molécule cible y soit immobilisée. Ensuite, (c) on génère un second spectre RMN des composés à soumettre à recherche systématique en présence du support solide sur lequel est immobilisée la molécule cible. Enfin, (d), on compare les deux spectres à la recherche des différences entre eux. L'invention concerne enfin un appareil de RMN dans lequel est disposée une sonde RMN particulière.
