Disclosed herein is a biological wound dressing having good biocompatibility, durable flexibility, that is gas permeable but impenetrable to bacteria, that is convenient to use, and a method of preparation thereof. The biological wound dressing comprises a substrate prepared from animal tissue. The dressing is made by a method comprising: crosslinking and fixing the substrate with a non-aldehyde fixative; minimizing antigens from the substrate; and incorporating an active layer in the substrate. Minimizing the antigens from the substrate comprises: utilizing a first reagent selected from low molecular weight organic acid anhydride, acyl chloride, acylamide, or monocyclic oxide, to block one or more antigen determinants of the substrate; and utilizing a second reagent, being a guanidine compound, to replace hydrogen bonding in the spiral chains of the protein molecules in the substrate and alter its specific conformation.
