Agents for making and breaking prions (PrP Sc) under "physiological" conditions, in vitro, are revealed. These include a synthetic copy of a domain of a bovine/ovine naturally occurring protein ("cf22aa") that converts constitutive native human brain prion proteins (PrPC) to (PrP Sc), the other, a synthetic copy of a domain of a protein encoded on the anti-sense strand of human P53 gene, ("pbf13aa") breaks-up PrP from TSE infected human brains, and PrP Sc artificially added to human blood and other fluids. Also revealed are two mono-specific antibodies which block conversion, to PrP Sc and remove excess of pbf13aa from fluids treated with this polypeptide, respectively.
