A modified hemoglobin comprising hemoglobin which is cross-linked with a cross-linking reagent. The cross-linking rea- gent comprises an aromatic- or aliphatic-derived acyl material having at least two anionic leaving groups, each of them being ad- jacent to an electrophile group. Such agent provides a cross-link between the beta-chains within the 2,3-diphosphoglycerate bind- ing site with a linkage distance between the beta-chains of about 5 to 9 angstroms. A method of preparing the modified hemoglobin and its use as a blood substitute or a plasma expander are also described.
