The present invention provides methods and compositions for expanding Treg cells ex vivo or in vivo using one or more conjugates comprising a costimulatory moiety that stimulates at least one of three signals involved in Treg cell development and/or using dendritic cells pulsed with antigens and modified to display TGF-.beta., or hematopoetic stem cells or bone marrow cells modified to display TGF-.beta.. The methods and compositions are useful, for example, in the treatment and prevention of autoimmune disease, including Type 1 diabetes and in preventing foreign graft rejection, as well as to establish mixed chimerism, induce tolerance to autoantigens, alloantigens or xenoantigens, beta cell regeneration, prevention of foreign graft rejection, and treatment of a genetically inherited hematopoietic disorder.
L'invention concerne des procédés et des compositions permettant d'étendre des lymphocytes Treg ex vivo ou in vivo au moyen d'un ou de plusieurs conjugués comprenant une fraction co-stimulatoire stimulant au moins un des trois signaux impliqués dans le développement des lymphocytes Treg et/ou au moyen de cellules dendritiques pulsée avec des antigènes et modifiées afin de présenter TGF-.beta. ou des cellules souches hématopoiétiques ou des cellules de la moelle osseuse modifiées pour présenter TGF-.beta.. Les procédés et les compositions sont utiles, par exemple, dans le traitement et la prévention de maladie autoimmune, notamment les diabètes de Type 1 et la prévention du rejet du greffon étranger, ainsi que pour établir un chimérisme mixte, pour induire une tolérance à des auto-antigènes, des alloantigènes ou des xénoantigènes, une régénération des cellules bêta, la prévention du rejet de greffon étranger et le traitement d'un trouble hématopoiétique hérité sur le plan génétique.
