The present invention provides a novel dominant selectable marker system in yeast that is based on an aminoglycoside, nourseothricin (NST). This compound possesses a powerful antifungal activity against Candida albicans and S. cerevisiae. The invention provides a cognate drug resistance marker for use in gene transformation and disruption experimentation in Candida albicans and Saccharomyces cerevisiae. In particular, the invention presents: 1) direct utility for gene manipulations in both clinically and experimentally relevant strains regardless of genotype and without affecting growth rate, or hyphal formation; and 2) applicability to antifungal drug discovery, including target validation and various forms of drug screening assays.
La présente invention concerne un nouveau système de marqueur dominant sélectionnable dans la levure basé sur un aminoglycoside, nourseothricine (NST). Ledit composé présente une activité antifongique puissante vis-à-vis du Candida albicans et de S.cerevisiae. L'invention propose un marqueur apparenté de résistance aux médicaments utilisable dans la transformation des gènes et dans l'expérimentation de dissociation dans le Candida albicans et le Saccharomyces cerevisiae. En particulier, l'invention présente: 1) une aptitude directe pour les manipulations génétiques sur des souches appropriées sur le plan clinique ainsi que sur celui de l'expérimentation indépendamment du génotype et sans altération du taux de croissance, ou la formation fongique; et 2) une applicabilité à la découverte de médicaments antifongiques, y compris la validation des cibles et les diverses formes d'essais biologiques pour le criblage des médicaments.
