A process for the preparation of cholyl-L-lysine comprising the steps of:- (a) reacting N-.epsilon.-CBZ-cholyl-L-lysine with a hydrogen source in the presence of a catalyst in a solvent comprising one or more alkanols; (b) removing the catalyst; (c) optionally diluting the resulting reaction mixture with water and optionally adjusting the pH of the resultant reaction mixture to a pH less than or equal to about 4; (d) removing the bulk of the alkanol whilst ensuring that the alkanol content of the resultant mixture is maintained above about 3% w/w of the remaining mixture; (e) extracting the resultant mixture from step (d) with an organic solvent; (f) adjusting the pH of the aqueous layer to a pH of about 4.5 or greater to precipitate cholyl-L-lysine; (g) isolate the precipitate.

La présente invention a pour objet un procédé de préparation de cholyl-L-lysine comprenant les étapes consistant à : (a) faire réagir de la N-e-CBZ-cholyl-L-lysine avec une source d'hydrogène en présence d'un catalyseur dans un solvant comprenant un ou plusieurs alcanols ; (b) éliminer le catalyseur ; (c) diluer éventuellement le mélange réactionnel résultant avec de l'eau et ajuster éventuellement le pH du mélange réactionnel résultant à un pH inférieur ou égal à environ 4 ; (d) éliminer un gros volume de l'alcanol tout en s'assurant que la teneur en alcanol du mélange résultant est maintenue au-dessus d'environ 3 % p/p du mélange restant ; (e) extraire le mélange résultant issu de l'étape (d) avec un solvant organique ; (f) ajuster le pH de la couche aqueuse à un pH d'environ 4,5 ou supérieur pour précipiter la cholyl-L-lysine ; (g) isoler le précipité.
