Methods for producing genetically modified plants, particularly woody plants, and most particularly plants of the Eucapyptus and Pinus species, involve transformation of target plant material with a desired genetic construct and regeneration of the transformed plant material using an adventitious shoot bud system. The methods provide a high transformation efficiency and substantially reduce the duration of the transformation and regeneration protocols. Stem segments of a target plant are transformed using Agrobacterium-mediated techniques, and adventitious shoot buds are regenerated from the Agrobacterium- infected stem segments. Preferred culture media, including selection media, and improved plant culture techniques are disclosed.
L'invention concerne des procédés permettant de produire des plantes génétiquement modifiées, notamment des plantes ligneuses, et plus particulièrement des plantes des espèces Eucapyptus et Pinus. Ces procédés impliquent la transformation d'un matériau végétal cible en une structure génique désirée, et la régénération du matériau transformé au moyen d'un système de bourgeon adventif de pousse. Ces procédés présentent une efficacité de transformation très élevée, et réduisent sensiblement la durée des protocoles de transformation et de régénération. Des segments de tige d'une plante cible sont transformés à l'aide de techniques induites par Agrobactérium, et les bourgeons adventifs de pousse sont régénérés à partir de segments de tige infectés par Agrobacterium. L'invention concerne également un milieu de culture préféré comprenant un support de sélection, et une amélioration des techniques de culture des plantes.
