Biological selection systems of the invention provide methods for selecting a pathway modulating protein that produces a desired target moiety that acts as a transcriptional effector. The system includes a library of expressible coding sequences where each expressible coding sequence encodes a variant of the pathway modulating protein, such that the variants of the pathway modulating protein differ in their ability to produce the target moiety. The system further comprises a host expression system for expressing the library of expressible coding sequences, wherein each host expression system expresses a coding sequence from the library. Each host expression system includes a viability modulating promoter, wherein expression mediated by the viability modulating promoter determines the survival of the host expression system. Each host expression system also includes an expressed target-moiety-responsive transcription factor, wherein the transcription factor is capable of mediating expression by the viability modulating promoter. The transcription factor responds to binding the target moiety by mediating expression at the viability modulating promoter. Host systems expressing a selected pathway modulating protein so as to produce the desired target moiety may thereby be selected based on host viability.
