The invention relates to 2-hydroxytetrahydrofurane derivatives having general formula (I) wherein A represents the radical (II) wherein R?1¿, R?2¿, R?4¿, R?5¿ and R?6¿ individually represent H, a halogen atom, OH, alkyl or alkoxy, R?3¿ represents H, alkyl or -COR?1()¿, R?10¿ represents H, alkyl or alkoxy, and W represents a CH¿2?-CH¿2?-, -CH=CH-, -0-, -S- or -NR?11 ¿bond? ¿wherein R?11¿ represents H or alkyl; X represents CO-, -Y-CO-, -O-Y-CO- or -NR?12¿-Y-CO-, Y represents an alkylene radical or a haloalkylene radical, R?12 ¿represents H, alkyl or -COR?13¿¿,? R?13¿ represents H, alkyl, haloalkyl or alkoxy, AA represents, each time when it occurs, a natural amino acid or not; n represents 2 or 3; and R represents H, alkyle or -CO-R?19¿, R?19 ¿represents alkyl. Said compounds inhibit calpains and / or trap reactive forms of oxygen and can be used to prepare a medicament used to treat inflammatory diseases and immunological diseases, cardiovascular diseases and cerebro-vascular diseases, disorders of the central or peripheral nervous system, osteoporosis, muscular distrophy, proliferative diseases, cataracts, rejection reactions following organ transplants and viral and auto-immune diseases.
L'invention concerne des dérivés du 2-hydroxytétrahydrofuranne répondant à la formule générale (I) dans laquelle A représente le radical (II) dans lequel R1, R2, R4, R5 et R6 représentent (notamment), indépendamment, H, un atome halogène, OH, alkyle ou alkoxy, R3 représente H, alkyle ou -COR1(), R10 représentant H, alkyle ou alkoxy, et W représente une liaison, -CH2-CH2-, - CH=CH-, -0-, -S- ou -NR11- dans lequel R11 représente H ou alkyle ; X représente -CO-, -Y-CO-, -O-Y-CO- ou -NR12-Y-CO-, Y représente un radical alkylène ou haloalkylène, R12représente H, alkyle ou -COR13, R13 représente H, alkyle, haloalkyle ou alkoxy, AA représente, à chaque fois qu'il intervient, un aminoacide naturel ou non ; n représente 2 ou 3 ; et enfin R représente H, alkyle ou -CO-R19, R19 représentant alkyle. Ces composés présentent une activité inhibitrice des calpaïnes et / ou une activité piégeuse des formes réactives de l'oxygène et peuvent être utilisés pour préparer un médicament destiné à traiter les maladies inflammatoires et immunologiques, les maladies cardio-vasculaires et cérébro-vasculaires, les troubles du système nerveux central ou périphérique, l'ostéoporose, les dystrophies musculaires, les maladies prolifératives, la cataracte, les réactions de rejet suite à des transplantations d'organes et les maladies auto-immunes et virales.
