New methods for the rational identification of molecules capable of interacting with specific naturally occurring proteins are provided, in order to yield new pharmacologically important compounds and treatment modalities. Broadly, the method comprises the steps of identifying a switch control ligand forming a part of a particular protein of interest, and also identifying a complemental switch control pocket forming a part of the protein and which interacts with said switch control ligand. The ligand interacts in vivo with Hie pocket to regulate the conformation and biological activity of the protein such that the protein assumes a first conformation and a first biological activity, upon the ligand-pocket interaction, and assumes a second, different conformation and biological activity in the absence of the ligand-pocket interaction. Next, respective samples of said protein in the first and second conformations are provided, and these are screened against one or more candidate molecules by contacting the molecules and the samples. Thereupon, small molecules which bind with the protein at the region of the pocket maybe identified. Novel protein-modulator adducts and methods of altering protein activity are also provided.
Méthodes d'identification rationnelle de molécules, permettant d'interagir avec des protéines naturelles spécifiques afin de mettre au point des nouveaux composants et modalités de traitement importants au plan pharmacologique. Schématiquement, cette méthode consiste à identifier un ligand de commande de changement d'état faisant partie d'une protéine particulière d'intérêt ainsi qu'une poche complémentaire de commande de changement d'état faisant partie de la protéine et interagissant avec ledit ligand. Ce ligand interagit in vivo avec la poche et régule la conformation et l'activité biologique de la protéine de sorte que cette protéine a une première conformation et une première activité biologique lors de l'interaction ligand-poche, et une seconde et différente conformation et activité en l'absence de ladite interaction. On prend ensuite des échantillons de ladite protéine dans la première et la seconde conformations, échantillons que l'on soumet à un criblage par rapport à une ou plusieurs molécules candidates, ceci par contact entre molécules et échantillons. On parvient ainsi à identifier des petites molécules qui se lient avec la protéine dans la région de la poche. Sont également décrits des nouveaux adduits st des procédés permettant de modifier l'activité protéique.
