The invention relates to methods and kits for separating, and optionally analyzing, substoichiometrically interacting ligands from a high abundance ligand of interest in an affinity matrix system. The methods comprising immobilizing a complex of a first and second ligand to an affinity matrix, wherein the second ligand is bound to the matrix, and subsequently eluting the first ligand from the matrix. In addition, the methods and kits can be used for drug discovery.
