A method of obtaining nuclei from target nucleated cells e.g. foetal cells, when these cells are in admixture with other nucleated cells e.g. in maternal blood, comprises treating the maternal blood with a lytic reagent so as to selectively lyse the plasma membranes but not the nuclear membranes of the target nucleated cells while leaving the other nucleated cells substantially intact, separating the target cell nuclei and analysing either the isolated target cell nuclei or the remaining intact cells.

La présente invention a trait à un procédé permettant d'obtenir des noyaux à partir de cellules nucléées cibles telles que les cellules foetales, lorsque ces cellules sont mélangées à d'autres cellules nucléées, par exemple, dans le sang maternel. Le procédé consiste à traiter le sang maternel avec un réactif lytique de façon à lyser sélectivement les membranes plasmiques et non les membranes nucléaires des cellules nucléées cibles, tout en conservant pratiquement intactes les autres cellules nucléées, à séparer les noyaux des cellules cibles et à analyser soit les noyaux des cellules cibles isolées soit les cellules restées intactes.
