The purification of native RB (retinoblastoma) as a complex, including P107, P130, and a 600 kDa subunit, termed MTAF600 (microtubule associated factor 600) is described. MTAF600 binds to RB regardless of the phosphorylation status of RB, and binds to RB without disrupting the interaction between RB and E2F. It is further shown that E2F and DP proteins co-purified with MTAF600 and RB, such that hypophosphorylated RB may gain access to E2F as a complex with MTAF600. In addition, MTAF600 binds to microtubules and plays a role in active repression of E2F-responsive genes, cell cycle arrest, and genomic stability. The sequence of MTAF600 is described herein, along with its binding properties to proteins such as RB and microtubules, and its sequence homology. Further, methods and reagents for assaying the presence of MTAF600 or mutants thereof, pharmaceutical formulations, and methods for treating disease are also described.
L'invention porte sur la purification de RB (rétinoblastome) présent dans un complexe comportant des P107, P130, et une sous-unité de 600 kDa, le MTAF600 (facteur 600 associé aux microtubules). Ledit MTAF600 se fixe au RB indépendamment de son état de phosphorylation et sans interrompre l'interaction entre le RB et l'E2F. Il est par ailleurs démontré que les protéines E2F et DP, co-purifiées avec le MTAF600 et le RB, permettent au RB hypophosphorylé d'avoir accès à l'E2F dans le complexe comprenant le MTAF600. De plus, le MTAF600 se fixe aux microtubules et joue un rôle actif dans la répression des gènes répondant à l'E2F, dans l'arrêt du cycle cellulaire, et dans la stabilité du génome. La séquence du MTAF600 et son homologie sont décrites dans la description, ainsi que leurs propriétés de fixation à des protéines telles que le RB, et aux microtubules, et à leurs séquences homologues. L'invention porte en outre sur des procédés et réactifs de détection de la présence du MTAF600 ou de ses mutants, sur des préparations pharmaceutiques, et sur des procédés de traitement de maladies.
