The invention is a method of identifying the presence of, and monitoring, a disease state in a mammal which is associated with degradation of connective tissue in the mammal. The method detects and determines whether diagnostically or prognostically significant levels of YKL-40 protein and/or YKL-40 peptide are present in a biological sample. The method can be used, for example, to identify the presence of inflammatory or degenerative joint desease or degeneration of connective tissue in organs. Serum YKL-4O levels as detected and quantified by the inventive method are also suggestive of the prognosis for the length of survival in breast cancer patients following recurrence and/or metastasis of weir cancers. The figure shows the elution position of substantially pure serum YKL-40 on a gel filtration column.
Procédé permettant d'identifier la présence, chez un mammifère, d'un état pathologique associé à la dégradation de tissus conjonctifs, et de surveiller cet état. Ledit procédé permet de détecter et de déterminer, à des fins diagnostiques ou pronostiques, si une protéine YKL-40 et/ou un peptide YKL-40 sont présents en quantité importante dans un échantillon biologique. Ledit procédé peut être utilisé, par exemple, pour identifier la présence de maladies inflammatoires ou dégénératives des articulations, ou la dégénérescence de tissus conjonctifs dans des organes. Les niveaux de YKL-40 sérique détectés et quantifiés selon le procédé de la présente invention permettent également d'établir un pronostic sur la durée de survie de patientes atteintes de cancer du sein à la suite d'une rechute et/ou de l'apparition de métastases de leur cancer. La figure montre la position d'élution de YKL-40 sérique sensiblement pure sur une colonne de filtration sur gel.
