The invention relates to an assay for testing oxidative stress of a subject by measurement of oxidants in biological fluids such as urine, plasma, bioreactor medium and respiratory aspirants. There is provided a method of determining oxidative stress in a mammalian subject. The method comprises: obtaining a sample of a biological fluid from the subject; mixing the biological fluid with a ferrous reaction reagent; incubating the biological fluid and the reaction reagent; and detecting a coloured reaction product. There is further provided a ferrous reaction reagent suitable for use in assaying oxidative stress, said reaction reagent comprising 2-deoxyglucose, TBA, EDTA, and ferrous sulfate, and being substantially free of ascorbic acid.
L'invention concerne un essai qui détermine le stress oxydatif induit dans un milieu en mesurant les oxydants présents dans des liquides biologiques comme l'urine, le plasma, le milieu d'un bioréacteur et les liquides aspirés dans les voies respiratoires. On y trouve une méthode permettant de déterminer le stress oxydatif chez un mammifère. La méthode comprend les étapes suivantes : obtenir un échantillon de liquide biologique provenant du mammifère, mélanger le liquide biologique avec un réactif ferreux, incuber le liquide biologique et le réactif de la réaction et détecter le produit de réaction coloré. L'invention porte aussi sur un réactif ferreux à utiliser pour mesurer le stress oxydatif, ledit réactif étant constitué de 2-désoxyglucose, de TBA, d'EDTA et de sulfate de fer, et étant essentiellement sans acide ascorbique.
