A novel method for the qualitative and quantitative analysis of dicarboxylic acids in biological samples is provided. The method includes the steps of esterifying the acid component of the sample and subsequently analyzing the esterified sample using tandem mass spectroscopy using atmospheric pressure ionization techniques in the positive ion mode. The method is particularly useful in the determination of methylmalonic acid in biological samples, and thus, the diagnosis of vitamin B12 deficiency.

L'invention concerne une nouvelle méthode destinée à l'analyse qualitative et quantitative des acides dicarboxyliques contenus dans les échantillons biologiques. La méthode comprend les étapes suivantes : estérification de la composante acide de l'échantillon, puis, analyse de l'échantillon estérifié au moyen de la spectrométrie de masse en tandem et à l'aide de techniques d'ionisation à pression atmosphérique dans le mode positif. La méthode est particulièrement utile pour mesurer l'acide méthylmalonique dans les échantillons biologiques, et par conséquent, pour diagnostiquer une carence en vitamine B12.
