An isolated nucleic acid molecule capable of expressing an human ether-à-go-go related gene (hERG) mutant that enhances hERG inactivation, wherein the mutation is not located in the vicinity of the drug-binding pocket within the cavity of the ion conduction pore of the hERG protein. Use of the isolated nucleic acid molecule in an assay for screening potential drug bind-ing to hERG protein.

Linvention concerne une molécule dacide nucléique isolée apte à exprimer un mutant de gène humain apparenté au gène éther-à-go-go (hERG) qui augmente linactivation de hERG, la mutation nétant pas située au voisinage de la poche de liaison à un médicament à lintérieur de la cavité du pore de conduction ionique de la protéine hERG. Linvention concerne également lutilisation de la molécule dacide nucléique isolée dans un essai pour le criblage dun médicament potentiel fixant la protéine hERG.
