Aggregation is a major cause of the misbehavior of proteins. A system for modifying a protein to create a more stable variant is provided. The method involves identifying non- conserved hydrophobic amino acid residues on the surface of a protein, suitable for mutating to more hydrophilic residues (e.g., charged amino acids). Any number of residues on the surface may be changed to create a variant that is more soluble, resistant to aggregation, has a greater ability to re-fold, and/or is more stable under a variety of conditions. The invention also provides GFP, streptavidin, and GST variants with an increased theoretical net charge created by the inventive technology. Kits are also provided for carrying out such modifications on any protein of interest.

L'agrégation est une cause majeure de dysfonctionnement des protéines. L'invention concerne un système permettant de modifier une protéine afin de créer un variant plus stable. Le procédé implique d'identifier à la surface d'une protéine des résidus acides aminés hydrophobes non conservés qui sont adéquats pour une mutation en résidus plus hydrophiles (c'est-à-dire en acides aminés chargés). Un nombre quelconque de résidus de la surface peut être modifié afin de créer un variant qui est plus soluble et résistant à l'agrégation, qui possède une plus grande capacité de repliement et/ou qui est plus stable dans des conditions diverses. L'invention concerne également des variants des protéines GFP, streptavidine et GST ayant une charge nette théorique plus élevée, créés par la technologie de l'invention. Elle propose également des kits pour réaliser ces modifications sur n'importe quelle protéine d'intérêt.
