A new method for DNA diagnostics based on template-directed primer extension and detection by fluorescence polarization is described. In this method, amplified genomic DNA fragments containing polymorphic sites are incubated with an oligonucleotide primer designated to hybridize to the DNA template adjacent to the polymorphic site in the presence of allelic dye-labeled dideoxyribonucleoside triphosphates and a modified Taq DNA polymerase. The primer is extended by the dye-terminator specific for the allele present on the template. At the end of the reaction, the fluorescence polarization of the two dye-terminators in the reaction mixture is analyzed directly without separation or purification. This homogeneous DNA diagnostic method is shown to be highly sensitive and specific and is suitable for automated genotyping of large number of samples.

L'invention concerne un nouveau procédé de diagnostic d'ADN fondé sur l'extension d'amorce dirigée sur le modèle et la détection par polarisation de fluorescence. Selon ce procédé, des fragments amplifiés d'ADN génomique contenant des sites polymorphes sont incubés au moyen d'une amorce polynucléotidique conçue pour s'hybrider avec le modèle d'ADN adjacent au site polymorphe en présence de triphosphates didésoxyribonucléosides alléliques marqués au colorant et d'une Taq ADN polymérase. L'amorce est étendue au moyen d'un terminateur de colorant spécifique à l'allèle présent dans le modèle. A la fin de la réaction, on analyse directement, sans séparation ni purification, la polarisation de fluorescence des deux terminateurs de colorant dans le mélange de réaction. Ce procédé homogène de diagnostic d'ADN s'avère très sensible et spécifique; il convient au génotypage automatique d'une grande quantité d'échantillons.
