Mismatch Repair Detection (MRD), a novel method for DNA-variation detection, utilizes bacteria to detect mismatches by a change in expression of a marker gene. DNA fragments to be screened for variation are cloned into two MRD plasmids, and bacteria are transformed with heteroduplexes of these constructs. Resulting colonies express the marker gene in the absence of a mismatch, and lack expression in the presence of a mismatch. MRD is capable of detecting a single mismatch within 10 kb of DNA. In addition, MRD can analyze many fragments simultaneously, offering a powerful method for high-throughput genotyping and mutation detection.

Cette invention se rapporte à une détection pour réparation de mésappariement (MRD), un nouveau procédé qui permet de détecter des variations d'ADN, et qui utilise des bactéries afin de détecter les mésappariement en se fondant sur un changement de l'expression d'un gène marqueur. Des fragments d'ADN, qui doivent être criblés afin de détecter une variation, sont clonés en deux plasmides de MRD, et des bactéries sont transformées à l'aide d'hétéroduplex des ces constructions. Les colonies résultantes expriment le gène marqueur en l'absence de mésappariement, et sont exemptes d'expression en présence d'un mésappariement. Cette MRD permet de détecter un seul mésappariement dans 10 kb d'ADN, et permet en outre d'analyser simultanément de nombreux fragments, offrant ainsi un procédé puissant en vue d'un génotypage et d'une détection de mutations à haut rendement.
