The use of a means to vary Ubc4p or Ubc5p activity in a fungal cell to control the copy number of a plasmid in the cell. The level of Ubc4p or Ubc5p activity may be reduced/abolished (for example by gene deletion, mutagenesis to provide a less active protein, production of antisense mRNA or production of competitive peptides) to raise the copy number and increase yield of a protein encoded by the plasmid.

Cette invention se rapporte à un procédé conçu pour faire varier l'activité Ubc4p ou Ubc5p d'une cellule fongique de manière à réguler le numéro de transcription d'un plasmide dans la cellule. Il est possible de réduire le niveau de l'activité Ubc4p ou Ubc5p ou de supprimer cette activité (par exemple par délétion de gènes, mutagenèse pour produire une protéine moins active, production d'ARNm antisens ou production de peptides concurrentes) de façon à augmenter le numéro de transcription et à accroître le rendement d'une protéine codée par le plasmide.
