The present invention relates to a method for producing a protein of interest using a fusion protein that includes a protein of interest as well as a marking domain consisting of a histidine sequence, a domain for interacting with streptavidin and a colored marker, the marking domain and the protein of interest being separated by a cleavage site specific to enterokinase or TEV protease. The invention further relates to a chromatography column for implementing the method of the present invention.

La présente invention concerne un procède de production d'une protéine d'intérêt grâce à l'utilisation d'une protéine de fusion qui comprend une protéine d´intérêt ainsi qu'un domaine de marquage constitue d'une séquence d'histidine, d'un domaine d' interaction a la streptavidine et d'un marqueur colore, le domaine de marquage et la protéine d' intérêt étant séparés par un site de clivage spécifique de l'entérokinase ou Ia TEV protéase. En outre, la présente invention concerne une colonne de chromatographie permettant la mise en oeuvre du procède selon la présente invention.
