An invention is described which allows measurement of the concentration of fluorescent particles (210) in a flowing (laminar) fluid by imaging the flow (214) with a video camera or the like. A beam of illumination is used to illuminate the target particles. Imaging optics (408, 412) are arranged to view the focal plane to form an image of the multiple fluorescent sample particles in the flow stream; a camera records the image formed by the imaging optics, and a counting algorithm (812) enumerates the particles. Operational parameters of the system are adjusted (824) according to an initial estimate of particle density (828), for example flow rate (824a), exposure time, and sampling interval (824b). In addition, the counting algorithm is selected according to the estimated particle density.
L'invention concerne la mesure de la concentration de particules fluorescentes (210) dans un écoulement de fluide (laminaire) consistant à imager l'écoulement (214) au moyen d'une caméra vidéo ou analogue. Un faisceau d'éclairage est utilisé pour éclairer les particules cibles. Des éléments optiques d'imagerie (408, 412) sont disposés de manière à visualiser le plan focal, de manière à former une image des particules fluorescentes multiples de l'échantillon dans l'écoulement; une caméra enregistre l'image formée par les éléments optiques d'imagerie et un algorithme de décompte (812) énumère les particules. Des paramètres fonctionnels du système sont réglés (824) en fonction d'une estimation initiale de la densité des particules (828), par exemple, le débit (824a), le temps d'exposition et l'intervalle d'échantillonnage (824b).
