A purified thermostable enzyme is derived from the archael bacterium Thermococcus GU5L5. The enzyme has a molecular weight of about 68.5 kilodaltons and has cellulase activity. The enzyme can be produced from native or recombinant host cells and can be used for the removal of arginine, phenylalanine, or methionine amino acids from the N-terminal end of peptides in peptide or peptidomimetic synthesis. The enzyme is selective for the L, or "natural" enantiomer of the amino acid derivatives and is therefore useful for the production of optically active compounds. These reactions can be performed in the presence of the chemically more reactive ester functionality, a step which is very difficult to achieve with nonenzymatic methods.

Enzyme thermostable purifiée provenant de l'archaeobactérie Thermococcus GU5L5. Cette enzyme possède un poids moléculaire de 68,5 kilodaltons, ainsi qu'une activité cellulaire. Elle peut être obtenue à partir de cellules hôtes natives ou de cellules hôtes de recombinaison et utilisée afin de supprimer les acides aminés d'arginine, de phénylalanine ou de méthionine depuis l'extrémité de N-terminaison de peptides dans la synthèse de peptides ou de mimétiques de peptides. Cette enzyme présente une sélectivité pour l'énantiomère L ou "naturel" des dérivés d'acides aminés et est, de ce fait, utile pour la préparation de composés efficaces sur le plan optique. On peut effectuer ces réactions en présence de la valence fonctionnelle d'ester présentant une réactivité plus importante sur le plan chimique, cette étape étant très difficile à réaliser avec des procédés non enzymatiques.
