The invention is related to a transducing particle that comprises a bacteriophage coat and a DNA core that comprises plasmid DNA comprising: a) a host-specific bacteriophage packaging site wherein the packaging site is substantially in isolation from sequences naturally occurring adjacent thereto in the bacteriophage genome, b) a reporter gene, c) a bacteria-specific promoter operably linked to said reporter gene, d) a bacteria-~specific origin of replication, and optionally e) an antibiotic resistance gene. The invention includes phage transducing particles, methods of making transducing particles, and methods of using the transducing particles in bacterial detection.
La présente invention concerne une particule transductrice qui comprend une enveloppe bactériophage et un noyau d'ADN qui comprend de l'ADN plasmidique comprenant: a) un site d'encapsidation de bactériophage spécifique à l'hôte tel que le site d'encapsidation est sensiblement isolé des séquences naturellement situées à côté de ce dernier dans le génome du bactériophage, b) un gène rapporteur, c) un promoteur spécifique aux bactéries fonctionnellement lié audit gène rapporteur, d) une origine de réplication spécifique aux bactéries et facultativement e) un gène de résistance aux antibiotiques. Cette invention concerne des particules transductrices de phage, des procédés de préparation de particules transductrices et des procédés d'utilisation desdites particules transductrices pour la détection de bactéries.
