A method of protein engineering is provided wherein a searchable computer database is created comprising entries in the form of descriptions of a location and orientation in 3D space of side chains of the constituent amino acid residues of a framework protein. A query is created which corresponds to a description of a location and orientation in 3D space of respective side chains of amino acid residues of a sample protein. The location and orientation in 3D space of constituent side-chains is preferably described as a C.alpha.-C.beta. vector. The query is used to search the database and thereby identify a hit which corresponds to a framework protein having structural similarity with said sample protein. Framework protein "hits" so identified may be suitable candidates for further modification. A particular advantage of the present invention is that a modified framework protein may display one or more desired characteristics, such as a function similar to or inhibitory of the sample protein.
L'invention porte sur un procédé visant à fabriquer des protéines. Pour cela, on crée une base de données informatique de recherche comprenant des entrées sous forme de descriptions d'un emplacement et d'une orientation dans un espace tridimensionnel de chaînes latérales des restes d'acides aminés constituants d'une protéine de structure. On formule une requête qui correspond à une description d'un emplacement et d'une orientation dans un espace tridimensionnel de chaînes latérales respectives des restes d'acides aminés d'une protéine échantillon. L'emplacement et l'orientation dans l'espace tridimensionnel des chaînes latérales constituantes sont de préférence décrits sous forme d'un vecteur C.alpha.-C.beta.. La requête sert à consulter la base de données et à identifier ainsi une réponse pertinente qui correspond à une protéine de structure ayant une similarité structurale avec la protéine échantillon. Les <= réponses pertinentes >= correspondant à la protéine de structure ainsi identifiées peuvent être retenues en vue d'une modification ultérieure. L'avantage spécifique de cette invention est qu'une protéine de structure modifiée peut présenter une ou plusieurs caractéristiques désirées telles qu'une fonction similaire ou inhibitrice de la protéine échantillon.
