A method is provided for producing a population of post-mitotic cells of the neutrophil lineage, which method comprises the ex vivo steps of: (a) providing a population of cells comprising neutrophil progenitor cells; and (b) culturing the population of cells in an animal cell culture medium comprising (i) one or more early acting cytokines and (ii) one or more cytokines that differentiate said progenitor cells into a neutrophil specific lineage, under conditions of low oxidative stress, the culture medium being agitated when the cells are at a cell density at which oxygen transfer via the surface of the culture medium is insufficient for growth of the progenitor cells and the progeny thereof under static conditions, to produce a population of post-mitotic cells of the neutrophil lineage. The resulting population of cells can be used to increase the number of neutrophils in a patient.
La présente invention concerne un procédé qui permet de produire une population de cellules post-mitotiques de lignée neutrophile. Ledit procédé comprend les étapes ex vivo consistant à : (a) disposer d'une population de cellules comprenant des cellules progénitrices neutrophiles ; et (b) cultiver la population de cellules dans un milieu de culture de cellules animales contenant (i) au moins une cytokine à l'activité précoce et (ii) au moins une cytokine qui différencie lesdites cellules progénitrices en une lignée neutrophile spécifique, dans des conditions de faible stress oxydatif. Lorsque les cellules possèdent une densité cellulaire rendant le transfert d'oxygène par la surface du milieu de culture insuffisant pour la croissance des cellules progénitrices et de sa descendance dans des conditions statiques, le milieu de culture est alors agité afin de produire des cellules post-mitotiques de lignée neutrophile. La population de cellules ainsi obtenue peut servir à augmenter le nombre de neutrophiles chez un patient.
