The invention relates to an autofluorescent fusion protein which is suitable for use as a protease substrate. The invention further relates to a nucleic acid sequence coding for said fusion protein, in addition to a method using the fusion protein and/or the nucleic acid sequence in a two coloured, confocal fluorometric assay for the detection and quantification of proteolytic activity in liquid samples or cells.
