The present invention relates to a process for preparing anthrax protective antigen protein from E.coli using fed batch culture. This process creates a constitutively expressing system for rapid, efficient, cost-effective and high- level production of anthrax PA from E.coli. The steps of the process involves, transforming E.coli DH5.alpha. cells with the recombinant constitutive expression plasmid containing the PA gene to obtain recombinant DH5.alpha. cells and testing the PA expression by lysis of said cells followed by denaturing gel electrophoresis and Western Blotting technique using PA antibodies. This is followed by fermentation and harvesting of the high cell density cells. The said cells are solubilized using 6-8 Molar Urea and separated by centrifugation. The urea denatured PA is isolated from said supernatant and purified and thereafter eluted.
L'invention concerne un procédé permettant de préparer une protéine d'antigène protecteur contre la maladie du charbon à partir de E.coli, au moyen d'une culture à écoulement discontinu. Le procédé selon l'invention permet de créer un système d'expression constitutive permettant la production rapide, efficace, rentable et à haut niveau d'un antigène protecteur contre la maladie du charbon, à partir d'E.coli. Ce procédé consiste à transformer des cellules DH5.alpha. de E.coli au moyen du plasmide à expression constitutive recombiné contenant le gène de l'antigène protecteur (AP), de sorte à obtenir des cellules DH5.alpha. recombinées; et à tester l'expression de l'AP par la lyse desdites cellules, puis par une électrophorèse sur gel de dénaturation et une technique de transfert Western utilisant des anticorps d'AP. Cette étape est suivie par la fermentation et la récolte des cellules à haute densité cellulaire. Lesdites cellules sont solubilisées au moyen d'une urée molaire 6-8 et séparées par centrifugation. L'antigène protecteur dénaturé par l'urée est isolé dudit surnageant, purifié, puis élué.

Very useful and original work well ahead of its peers meant to be a first E coli based recombinant anthrax vaccine up-scaled to high levels of production.
