Described herein is a novel expression system for producing multiple gene products of interest from a single polycistronic construct. In particular, the expression system contains a polycistronic vector capable of expressing functional antibodies in eukaryotic host cells, which vector contains at least the following elements operably linked in the 5' to 3' orientation: a promoter operable in a eukaryotic cell; a DNA sequence encoding at least the variable region of an antibody light chain; an internal ribosome entry site (IRES); and at least one DNA sequence encoding an antibody heavy chain. Also disclosed are mammalian cells containing the polycistronic expression vector, and a method of producing functional antibodies in mammalian cells transfected with the polycistronic expression vector.
L'invention concerne un nouveau système d'expression permettant de produire plusieurs produits géniques recherchés à partir d'une construction polycistronique unique. Ce système d'expression contient en particulier un vecteur polycistronique pouvant exprimer des anticorps fonctionnels dans des cellules hôtes eucaryotes, ledit vecteur contenant au moins les éléments suivants fonctionnellement liés dans l'orientation 5'-3': un promoteur utilisable dans une cellule eucaryote; une séquence ADN codant au moins la région variable d'une chaîne légère d'un anticorps; un site interne d'entrée du ribosome (IRES); et au moins une séquence ADN codant une chaîne lourde d'un anticorps. L'invention concerne également des cellules mammaliennes contenant ledit vecteur d'expression polycistronique, et un procédé de production d'anticorps fonctionnels dans des cellules mammaliennes transfectées avec ledit vecteur d'expression polycistronique.
