The present invention comprises a method for producing mammalian therapeutics free from prion contamination and cells for use in such methods. Such therapeutics are produced in somatic cells having a genome with an artificially altered PrP gene. The PrP gene in these cells may be ablated, or replaced by an exogenous inducible form of the PrP gene. The endogenous gene in the host cells may be disrupted, or disrupted and replaced by an exogenous PrP gene.

La présente invention concerne une méthode permettant de produire des agents thérapeutiques mammifères exempts d'une quelconque contamination provoquée par des prions, ainsi que des cellules destinées à être utilisées selon ces méthodes. Les agents thérapeutiques susmentionnés sont produits dans des cellules somatiques comprenant un génome avec un gène PrP artificiellement modifié. Le gène PrP de ces cellules peut être retiré, ou remplacé par une forme inductible exogène de ce gène PrP. Le gène endogène des cellules hôtes peut être soit dissocié, soit dissocié puis remplacé par un gène PrP exogène.
