This invention provides an isolated nucleic acid molecule encoding a Tumor necrosis factor Receptor-Associated Factor (TRAF) protein-interacting hereditary multiple extoses (TREX) protein. This invention also provides vectors comprising the isolated nucleic acid molecule encoding a TREX protein. This invention further provides a purified TREX protein and antibodies thereto. This invention provides oligonucleotides comprising a nucleic acid molecule of at least 15 nucleotides capable of specifically hybridizing with a unique sequence included within the sequence of an isolated nucleic acid molecule encoding TREX protein. This invention provides an antisense oligonucleotide comprising a sequence capable of specifically hybridizing with a unique sequence included within a genomic DNA molecule encoding a Tumor necrosis factor Receptor-Associated Factor (TRAF) protein-interacting hereditary multiple extoses (TREX) protein. This invention provides a monoclonal antibody directed to an epitope of a TREX protein. This invention provides methods of inhibiting TREX protein interaction with a TRAF protein; of inhibiting overexpression of TREX protein; of inhibiting growth of a tumor; of treating abnormalities in a subject associated with overexpression of TREX. This invention provides pharmaceutical compositions comprising oligonucleotides effective to prevent overexpression of a TREX protein or antibodies effective to block binding of a TREX protein to a TRAF protein; screening for compounds which inhibit TREX protein and TRAF protein binding; of detecting predispositions to cancers comprising TREX mutations; and of diagnosing cancer comprising TREX mutations.
Cette invention concerne une molécule d'acide nucléique isolée codant une protéine (TREX) à extoses mutiples héréditaires en interaction avec une protéine (TRAF) de facteur associé au récepteur du facteur de nécrose tumorale. Cette invention concerne également des vecteurs comprenant la molécule d'acide nucléique isolée codant une protéine TREX. L'invention concerne également une protéine TREX purifiée et des anticorps contre celle-ci. Cette invention a trait à des oligonucléotides comprenant une molécule d'acide nucléique d'au moins 15 nucléotides capable de s'hybrider spécifiquement à une séquence unique contenue dans la séquence d'une molécule d'acide nucléique isolée codant la protéine TREX. De plus, cette invention concerne un oligonucléotide antisens comprenant une séquence capable de s'hybrider spécifiquement à une séquence unique contenue dans une molécule d'ADN génomique codant une protéine (TREX) à extoses multiples héréditaires en interaction avec une protéine (TRAF) de facteur associé au récepteur du facteur de nécrose tumorale. Cette invention a également trait à un anticorps monoclonal dirigé contre un épitope d'une protéine TREX. Cette invention concerne également des méthodes d'inhibition de l'interaction de la protéine TREX avec une protéine TRAF, d'inhibition de la surexpression de la protéine TREX, d'inhibition de la croissance d'une tumeur, de traitement d'anomalies chez un sujet associé à une surexpression de TREX. L'invention concerne en outre des compositions pharmaceutiques contenant des oligonucléotides efficaces pour prévenir la surexpression d'une protéine TREX ou d'anticorps efficaces pour bloquer la liaison d'une protéine TREX à une protéine TRAF, pour cribler des composés inhibant la liaison de la protéine TREX et de la la protéine TRAF, pour détecter des prédispositions à des cancers comprenant des mutations TREX et pour diagnostiquer un cancer présentant des mutations TREX.
