A method is described for separating and purifying a wide variety of fermentation cyclopeptide products containing at least one protonatable amino group (including the deacylated Echinocandin-type compounds) from their fermentation or mixed broths and partially purified process streams by adsorbing the mixture onto a hydrophobic, reversed phase chromatographic media and eluting with a continuous linear acetic acid gradient ranging from 0.1 % acetic acid to 10.0 % acetic acid by volume in water. A process for removing tripeptide-aldehyde by-products from the fermentation products by means of a derivatizing agent is also described.
L'invention concerne un procédé de séparation et de purification s'appliquant à une large variété de produits cyclopeptidiques de fermentation contenant au moins un groupe amino protonable (tels que les composés de type Echinocandine désacylés) à partir de leur bouillons de fermentation ou mixtes et de flux de traitement partiellement purifiés. A cet effet, on adsorbe le mélange sur un support chromatographique hydrophobe en phase inverse puis on procède à son élution selon un gradient continu et linéaire correspondant à des concentrations d'acide acétique allant de 0,1 % à 10 %ar volume dans l'eau. L'invention concerne également un procédé permettant d'éliminer les sous-produits tripeptide-aldéhyde de ces produits de fermentation à l'aide d'un agent dérivatisant.
