The present invention provides an HSV having a genome from which, in the presence of the ICP4 gene product, a native immediate early gene is expressed with delayed kinetics, and an HSV having a genome with a mutation in each of the genes encoding ICP4, ICP27, and another HSV gene; preferably such HSV have one or more exogenous genes. The present invention further provides a method of expressing a polynucleotide within a cell comprising infecting the cell with such an HSV. Furthermore, the present invention provides a cell line having DNA encoding the HSV proteins ICP4, ICP27, and ICP0, and a method of producing an HSV vector by employing such a cell line.
La présente invention concerne un virus de l'herpès possédant un génome à partir duquel et en présence d'un produit génique ICP4 (peptide cellulaire infecté), un gène précoce, immédiat et natif est exprimé à l'aide de cinétiques différées, ainsi qu'un virus de l'herpès possédant un génome présentant une mutation dans chacun des gènes codant ICP4, ICP27, et un autre gène du virus de l'herpès, de tels virus possédant, de préférence, un ou plusieurs gènes exogènes. La présente invention concerne en outre un procédé d'expression d'un polynucléotide dans une cellule, consistant à infecter cette cellule au moyen d'un tel virus. En outre, la présente invention concerne encore une lignée cellulaire dont l'ADN code les protéines ICP4, ICP27 et ICP0 du virus de l'herpès, ainsi qu'un procédé de production d'un vecteur du virus de l'herpès, consistant à employer une telle lignée cellulaire.
