The present invention provides compounds, methods and systems for sequencing nucleic acid using single molecule detection. Using labeled NPs that exhibit charge-switching behavior, single-molecule DNA sequencing in a microchannel sorting system is realized. In operation, sequencing products are detected enabling real-time sequencing as successive detectable moieties flow through a detection channel. By electrically sorting charged molecules, the cleaved product molecules are detected in isolation without interference from unincorporated NPs and without illuminating the polymerase-DNA complex.

L'invention concerne des composés, des procédés et des systèmes destinés au séquençage d'acides nucléiques utilisant une détection de molécule unique. L'utilisation de phosphonucléotides marqués possédant un comportement de commutation de charges, rend possible la réalisation d'un séquençage d'une seule molécule d'ADN dans un système de séparation à microcanal. En fonctionnement, les produits de séquençage sont détectés, permettant un séquençage en temps réel lors du passage d'entités détectables successives dans un canal de détection. La séparation électrique de molécules chargées permet de détecter les produits de molécules fragmentées de manière isolée sans interférence qui pourrait provenir de nucléotides phosphates non incorporés et sans éclairage du complexe ADN-polymérase.
