ABSTRACT OF THE DISCLOSURE A method of preparing of N-glycosyl derivatives of polyene macrolides and salts thereof, particularly N-methylglucamine salts, which comprises: (a) reacting an amino group in the polyene macrolide with an aldose or ketose mono- or oligosaccharide in an organic solvent or mixture of organic solvents; (b) precipitating the thus formed N-glycosyl derivative from the reaction medium by the addition thereto of water or an aqueous solution of an inorganic salt, (c) recovering the thus obtained precipitate, (d) purifying the precipitate by recrystallization from an alkanol containing at least four carbon atoms, and (e) if desired converting the purified N-glycosyl derivative into a salt, and purifying this salt by a second recrystallization from an alkanol containing at least four carbon atoms. These derivatives show both enhanced activity and less toxicity anti-fungal agents, when compared to known soluble forms of polyene macrolide antibiotics such as the sodium desoxycholate complex of amphotericin B.
