A B S T R A C T The invention relates to a process for counting in vitro leucocytes, more particularly non degranulated basophils contained in a blood sample which comprises contact of the blood sample to be tested with a reagent containing a metachromate agent, a fixative fox leucocytes and basophils, and an acid present in such proportions that the final pH of the sample/reagent mixture is between about 3 and 5. Said process can be used for diagnosing in vitro anaphylactic sensitivity more particularly allergies.
