ABSTRACT OF THE DISCLOSURE Prothrombin in biological material, for example, blood plasma, is determined as a clinical parameter in the continuous monitoring of anticoagulation therapy by conversion of prothrombin in a sample of the material to thrombin by addition of Factor Xa, enzymatic fission of a thrombin substrate, for example, a peptide p-nitroanilide, measurement of a fission product, for example, nitroaniline. A novel reagent is also provided as well as a process for producing Factor Xa.
