The present invention is directed to substituted N-heterocycle derivatives of formula (I), compositions containing these derivatives, and their use in the manufacture of a medicament for the prevention and treatment of conditions ameliorated by monoamine reuptake including, inter alia, vasomotor symptoms (VMS), sexual dysfunction, gastrointestinal and genitourinary disorders, chronic fatigue syndrome, fibromylagia syndrome, nervous system disorders, and combinations thereof, particularly those conditions selected from the group consisting of major depressive disorder, vasomotor symptoms, stress and urge urinary incontinence, fibromyalgia, pain, diabetic neuropathy, and combinations thereof wherein : A is phenyl, naphthyl, thiophenyl, pyridinyl, furanyl, benzofuranyl, isobenzofuranyl, xanthenyl, pyrrolyl, indolizinyl, isoindolyl, indolyl, or benzothiophenyl, where 1 to 3 carbon atoms of said A is optionally replaced with a nitrogen atom; U is N or O; W is H or OR8; the other substituents are defined in the claims.
La présente invention porte sur des dérivés de N-hétérocycle substitués de formule (I), sur des compositions contenant ces dérivés et sur leur utilisation dans la fabrication d'un médicament utile dans la prévention et le traitement d'états améliorés par le recaptage de la monoamine tels, en autre, que les symptômes vasomoteurs, la dysfonction sexuelle, les troubles gastro-intestinaux et génito-urinaires, le syndrome de la fatigue chronique, le syndrome de la fibromyalgie, les troubles du système nerveux et des combinaisons de ceux-ci, notamment des états sélectionnés dans le groupe comprenant le trouble dépressif majeur, les symptômes vasomoteurs, le stress et l'incontinence urinaire impérieuse, la fibromyalgie, la douleur, la neuropathie diabétique et des combinaisons de ceux-ci. Dans cette formule: A représente phényle, naphthyle, thiophényle, pyridinyle, furanyle, benzofuranyle, isobenzofuranyle, xanthényle, pyrrolyle, indolizinyle, isoindolyle, indolyle ou benzothiophényle, les 1 à 3 atomes de carbone de A étant éventuellement remplacés par un atome d'azote; U représente N ou O; W représente H ou OR?8¿; les autres substituants étant définis dans les revendications.
