The present invention provides a class of compounds having the structure (see fig. I) or a pharmaceutically acceptable salt where R1 is hydrogen, hydroxy, alkoxy, alkoxycarbonyl, alkoxycarbonyloxy, aryloxycarbonyl, phenoxycarbonyloxy, or alkylsulfonyloxy; R2 and R3 are independently selected from hydrogen, halo, hydroxy, alkoxy, alkoxycarbonyl, alkoxycarbonyloxy, aryloxycarbonyl, phenoxycarbonyloxy, or alkylsulfonyloxy; R4 is selected from hydroxy, alkoxy, cycloalklxy, 1-piperidinyl, 1-pyrrolidinyl, methyl-1-pyrrolidinyl, dimethyl-1-pyrrolidino, 4-morpholino, dimethylamino, diethylamino, diisopropylamino, 1-hexamethyleneimino, and - OAr where where Ar is unsubstituted or substituted phenyl; X is selected from the group consisting of alkylene of two to four carbon atoms, -CH=CH-, -CH2CH=CH-, and -CH2CH2CH=CH-; and Y is absent or is carbonyl, with the proviso that when Y is absent, R4 may not be hydroxy, alkoxy, cycloalkoxy or aryloxy. The compounds are selective estrogen receptor modulators (SERM~s) and are useful in the treatment of pathological conditions associated with estrogen deprivation or the abnormal response to endogenous estrogen.

L'invention porte sur une classe de composés de structure (voir fig. I) ou aux sels acceptables en pharmacie, où R1 est un hydrogène ou un groupe hydroxy, alcoxy, alcoxycarbonyle, alcoxycarbonyloxy, aryloxycarbonyle, phénoxycarbonyloxy ou alkylsulfonyloxy; R2 et R3 sont indépendamment l'un de l'autre choisis parmi un hydrogène, un halogène ou un groupe hydroxy, alcoxy, alcoxycarbonyle, alcoxycarbonyloxy, aryloxycarbonyle, phénoxycarbonyloxy ou alkylsulfonyloxy; R4 est choisi parmi les groupes hydroxy, alcoxy, cycloalcoxy, 1-pipéridinyle, 1-pyrrolidinyle, méthyl-1-pyrrolidinyle, diméthyl-1-pyrrolidino, 4-morpholino, diméthylamino, diéthylamino, diisopropylamino, 1-hexaméthylèneimino, et -OAr, où Ar est un phényle avec ou sans substitution; X est choisi parmi un alkylène de deux à quatre atomes de carbone, -CH=CH-, -CH2CH=CH- et -CH2CH2CH=CH-; Y est un carbonyle ou absent, à la condition que, si Y est absent, R4 ne peut être un hydroxy, alcoxy, cycloalcoxy ou aryloxy. Ces composés, modulateurs sélectifs des récepteurs d'oestrogènes, sont utiles pour le traitement de troubles associés à la carence en oestrogènes ou a une réaction anormale aux oestrogènes endogènes.
