Abstract 1-N-Alkyl-substituted-kanamycin derivatives are prepared by a new process in which a 3",6'-di-N-acyl-3-N-formyl-kanamycin A or a 2',3",6'-tri-N- acyl-3-N-formyl-kanamycin B is alkylated, preferably by reductive alkylation. The process enables the alkyl kanamycin derivatives to be prepared from the readily available fermentation product kanamycin. The 3-N-formyl intermediates are new and their preparation is also described.
