The present invention relates to 11.beta.-fluoro-3-acetoxyestra-3,5-dien-17- one as a suitable intermediate in the preparation of 11-fluoro-subtituted steroids and to the process for preparation thereof. For this purpose, 11.alpha.-hydroxyestra-4-ene-3,17-dione is reacted with 1 to 3 equivalents of n-nonafluorobutanesulfonyl fluoride and 3 to 5 equivalents of diazabicycloundecene (DBU) at -40 to -20°C in an organic aprotic solvent and, after an aqueous workup, reacted with 5 to 10 equivalents of acetic anhydride and 0.01 to 1 equivalent of a strong acid. The desired product precipitates spontaneously out of the reaction solution and is obtained in a very high purity by filtration. The process is notable for the very high yield, avoidance of a chromatographic purification of the product, a reduced proportion of wastes and significantly increased process throughput. The process according to the invention is therefore especially suitable for preparing 11.beta.- fluoro-3-acetoxyestra-3,5- dien-17-one on a large industrial scale.
L'invention concerne du 11ß-fluoro-3-acétoxyestra-3,5-diène-17-one comme étape intermédiaire appropriée pour la fabrication de stéroïdes substitués fluoro-11, et son procédé de fabrication. À cet effet, du 11a-hydroxyestr-4-ène-3,17-dione est transformé, dans un solvant organique non protique, avec 1 à 3 équivalents de fluorure n-nonafluorobutane-sulfonyle et 3 à 5 équivalents de diazabicycloundécène (DBU) à -40 à -20 °C et, après un retraitement aqueux, est mélangé avec 5 à 10 équivalents d'anhydride acétique et 0,01 à 1 équivalent d'un acide concentré. Le produit souhaité se précipite spontanément à partir de la solution réactive, et est obtenu par filtration avec une très grande pureté. Le procédé se caractérise par un rendement très élevé, l'inutilité d'un nettoyage chromatographique du produit, une proportion réduite de déchets et un débit de traitement nettement accru. Le procédé selon l'invention convient donc notamment pour la fabrication de 11ß-fluoro-3-acétoxyestra-3,5-diène-17-one à grande échelle industrielle.
