The invention relates to benzoxazepinone derivatives of formula (I) wherein R1 is hydrogen, lower alkoxy, halogen or -NR'R"; n is 1 or 2; R',R" are independently from each other hydrogen or lower alkyl; R2 is hydrogen, lower alkyl, -(CH2)m-cycloalkyl, -(CH2)m-phenyl or -(CH2)m-O-lower alkyl; m is 0,1or 2; R3 is lower alkyl, -(CH2)m-C(O)O-lower alkyl, cycloalkyl or -(CH2)m-phenyl, which is unsusbtituted or substituted by one or two substituents, selected from the group consisting of halogen or lower alkyl; R4 is -(CH2).omicron.- phenyl, which is unsusbtituted or substituted by one or two substituents, selected from the group consisting of halogen, trifluoromethyl, -NR'R", nitro or_SO2NH2, or is - cycloalkyl, unsubstituted or substituted by phenyl, or is - (CR'R").omicron.-heterocyclyl, selected from the group defined in claim 1 and o is 1 or 2; and to pharmaceutically suitable acid addition salts thereof. These compounds are good .gamma.-secretase inhibitors for the treatment of Alzheimer's disease.
L'invention se rapporte à des dérivés de benzoxazepinone de formule (I), dans laquelle R?1¿ désigne hydrogène, alcoxy inférieur, halogène ou -NR"; n désigne 1 ou 2; N',R" sont pris indépendamment dans l'hydrogène ou l'alkyle inférieur; R?2¿ désigne hydrogène, alkyle inférieur, -(CH¿2?)¿m?.-cycloalkyle, -(CH¿2?)¿m?-phényle ou -(CH¿2?)¿m?-O-alkyle inférieur; m désigne 0, 1 ou 2; R?3¿ désigne alkyle inférieur, -(CH¿2?)¿m?-C(O)O-alkyle inférieur, cycloalkyle ou -(CH¿2?)¿m-?phényls, qui est non substitué ou substitué par un ou deux substituants, sélectionnés dans le groupe constitué d'halogène ou alkyle inférieur ; R?4¿ désigne -(CH¿2?)¿o?-phényle, qui est non substitué ou substitué par un ou plusieurs substituants, sélectionnés dans le groupe constitué d'halogène, trifluorométhyle, -NR'R", nitro ou -S0¿2?NH¿2?, ou désigne cycloalkyle , non substitué ou substitué par phényle, ou désigne (CR"R")¿o?hétérocyclyle, sélectionné dans le groupe défini dans la revendication 1 et o désigne 1 ou 2 ; et à leurs sels d'addition à un acide pharmaceutiquement acceptable. Ces composés sont de bons inhibiteurs de .gamma.-secretase pour le traitement de la maladie d'Alzheimer.
