2-alkylidene-18,19-dinor-vitamin D compounds are disclosed as well as pharmaceutical uses for these compounds and methods of synthesizing these compounds. These compounds are characterized by low bone calcium mobilization activity and high intestinal calcium transport activity. This results in novel therapeutic agents for the treatment and prophylaxis of diseases where bone formation is desired, particularly osteoporosis, as well as autoimmune diseases such as multiple sclerosis, diabetes mellitus and lupus. These compounds also exhibit pronounced activity in arresting the proliferation of undifferentiated cells and inducing their differentiation to the monocyte thus evidencing use as an anti-cancer agent and for the treatment of skin diseases such as psoriasis. These compounds also increase both breaking strength and crushing strength of bones evidencing use in conjunction with bone replacement surgery such as hip and knee replacements.

L'invention concerne des composés de vitamine D 2-alkylidène-18,19-dinor, ainsi que les utilisations pharmaceutiques desdits composés et des méthodes de synthèse desdits composés. Ces composés se caractérisent par une faible activité de mobilisation du calcium osseux et une activité de transport du calcium intestinal élevée. On obtient ainsi de nouveaux agents thérapeutiques utilisés dans le traitement et la prophylaxie de maladies dans lesquelles la formation osseuse est nécessaire, en particulier l'ostéoporose, ainsi que les maladies immunes telles que la sclérose en plaques, le diabète sucré et le lupus. Ces composés présentent également une activité prononcée dans l'arrêt de la prolifération des cellules non différenciées, et l'induction de leur différenciation par rapport au monocyte, permettant ainsi leur utilisation en tant qu'agent anticancéreux et dans le traitement des maladies cutanées telles que le psoriasis. Ces composés augmentent également la résistance à la rupture et la résistance à la compression des os, permettant ainsi leur utilisation conjointement à la chirurgie de remplacement osseux, par exemple le remplacement de la hanche ou du genou.
