2' and 3'-Prodrugs of l', 2', 3' or 4'-branched .szlig.-D or .szlig.-L nucleosides, or their pharmaceutically acceptable salts and derivatives are described, which are useful in the prevention and treatment of Flaviviridae infections and other related conditions. These modified nucleosides provide superior results against flaviviruses and pestiviruses, including hepatitis C virus and viruses generally that replicate through an RNA dependent RNA reverse transcriptase. Compounds, compositions, methods and uses are provided for the treatment of Flaviviridae infection, including HCV infection, that include the administration of an effective amount of the prodrugs of the present invention, or their pharmaceutically acceptable salts or derivatives. These drugs may optionally be administered in combination or alteration with further anti-viral agents to prevent or treat Flaviviridae infections and other related conditions.
L'invention concerne des prom~dicaments 2' et 3' de nucl~osides .szlig.-D ou .szlig.-L ramifi~s l', 2', 3' ou 4', ou leurs sels et d~riv~s pharmaceutiquement acceptables, utiles dans la pr~vention et le traitement d'infection par les Flaviviridae et d'autres ~tats associ~s. Ces nucl~osides modifi~s permettent d'obtenir de meilleurs r~sultats contre des flavivirus et des pestivirus, notamment le virus de l'h~patite C et des virus qui se r~pliquent au moyen d'une transcriptase inverse ARN-d~pendante. L'invention concerne des compos~s, des compositions, des m~thodes et des utilisations destin~s au traitement de l'infection par les Flaviviridae, notamment l'infection par le virus de l'h~patite C, qui impliquent l'administration d'une dose efficace desdits prom~dicaments, ou de leurs sels et d~riv~s pharmaceutiquement acceptables. Ces m~dicaments peuvent Útre ~ventuellement administr~s en combinaison avec d'autres agents antiviraux ou par alt~ration de ceux-ci afin de pr~venir ou de traiter des infections par les Flaviviridae et d'autres ~tats associ~s.
